BACKGROUND

Class II Human Leukocyte Antigens (HLA) are surface molecules expressed by professional antigen-presenting cells (dendritic cells, macrophages, B-lymphocytes and activated T cells), as well as certain normal and neoplastic non- lymphoid cells. There are three major allelic series of classical, surface- expressed class II molecules: HLA-DR, HLA-DQ, and HLA-DP, each comprising two polymorphic, glycosylated polypeptide chains (α and β) and associating with distinct sets of small peptide antigens.


PRODUCT

1. Each vial contains the indicated amount of IgG (caprylic acid purified) in 0.1% gelatine and 0.05% NaN3.

2. Unpurified reagent is provided at the indicated amounts with 0.1% NaN3.

Centrifuge the vial prior to use.


SPECIFICITY

The murine monoclonal antibody MM-P-5 (IgG1) recognizes a framework epitope that is (i) shared by the β chains of HLA-DR, -DQ as well as - DP molecules, and (ii) is present on these molecules regardless of their native (surfaceexpressed), or denatured form.


STORAGE

Store frozen, avoid repeated freeze-thawing. Stable for 2 yrs.

RESEARCH USE

This antibody is for laboratory research use only, not for human or in-vivo use.


APPLICATIONS

Flow cytometry,   10-20 µg/ml.

Immunohistochemistry (fluorescence, enzymatic),  Acetone-fixed cryostatic sections and cytospins (5 µg/ml). Formalin-fixed, paraffin-embedded sections with no need for antigen retrieval procedures. Under these conditions an overnight incubation was found to provide the most consistent staining patterns (2-5 µg/ml).

Immunoprecipitation,  1 to 5 µg, using rabbit anti-mouse Ig and protein A-sepharose beads.

Western blotting,   1 µg/ml.
